The aim of this project is to assess the impact of abasic sites that accumulate in the genome as a consequence of DNA demethylation. To this end, we intend to develop a new reagent for their quantification. The question of how the enzymes involved in DNA demethylation and base excision repair interact at their sites of action will be addressed with novel crosslinkers that will enable interactome studies in complex mixtures. Finally, we will use the newly developed crosslinker technology to study the mechanism of nucleotide excision repair in more detail.
Development of chemical mass spectrometry tools for the investigation of BER and NER
