Real-time analysis of replicative DNA damage processing

We will use a newly developed system for the introduction of localised lesions in the yeast genome for a time-resolved analysis of DNA replication in the presence of damage. By means of chromatin immunoprecipitation coupled to qPCR and next-generation sequencing as well as live-cell imaging, we will examine the mutual influences of DNA damage bypass and chromatin dynamics, aiming for a mechanistic understanding of the functions of histone modifications and chromatin remodellers in this process. In addition, we aim to isolate the damaged locus at distinct stages of its processing for proteomic analysis in order to identify novel factors involved in the replication of damaged DNA.
